Not sure - this is typically a networking issue - firewalls, subnets etc that you have already checked. No changes that you may not be aware of - router firmware update (broadcasts can sometimes be an issue - check IPMG proxy/snooping and UPNP settings if your router has them)?
Does the log file shed any light? Have you tried the latest Serviio version? Can you see the Windows Media Player server (if its enabled for streaming) from any device? All I can suggest is to double check everything network related - if wired try wireless and visa versa (if you can).
